Mechanisims of asthma and allergic disease – 1081. Resolvin inhibits the cryopyrin/nalp3 inflammasome

چکیده
Background The inflammasome is a novel protein complex that stimulates caspase-1 activation to promote the processing and secretion of IL-1b, a pro-inflammatory cytokine, which is among the most biologically important inflammatory mediators in allergic airway diseases. Among the various types of inflammasomes, Cryopyrin/NALP3 has been suggested to be involved in sensing sterile stress response, extracellular ATP, danger-associated molecular patterns (DAMPs), and crystals. Recently inappropriate NALP3 inflammasome activity has been reported in various allergic airway diseases including asthma. Therefore, inhibitors of the NALP3 inflammasome offer considerable therapeutic promise. Recently Omega-3 fatty acid derivates termed resolvins have shown to alter the effects of pro-inflammatory cytokine storm seen in inflammatory diseases. However the ability of resolvins to modulate the effects of inflammasome activation has not been studied. We investigated whether resolvin treatment inhibits inflammasome activation and regulates the functional effects of inflammasome mediated by IL-1b secretion.

Glyburide inhibits the Cryopyrin/Nalp3 inflammasome
Inflammasomes activate caspase-1 for processing and secretion of the cytokines interleukin-1beta (IL-1beta) and IL-18. Cryopyrin/NALP3/NLRP3 is an essential component of inflammasomes triggered by microbial ligands, danger-associated molecular patterns (DAMPs), and crystals. Inappropriate Cryopyrin activity has been incriminated in the pathogenesis of gouty arthritis, Alzheimer's, and silicosis...
متن کاملIdentification of Bacterial Muramyl Dipeptide as Activator of the NALP3/Cryopyrin Inflammasome
Activation of caspase-1 and subsequent processing and secretion of the pro-inflammatory cytokine IL-1beta is triggered upon assembly of the inflammasome complex. It is generally believed that bacterial lipopolysaccharides (LPS) are activators of the inflammasome through stimulation of Toll-like receptor 4 (TLR4). Like TLRs, NALP3/Cryopyrin, which is a key component of the inflammasome, contains...
متن کاملCryopyrin/NALP3 binds ATP/dATP, is an ATPase, and requires ATP binding to mediate inflammatory signaling.
The CATERPILLER (CLR/NLR) gene family encodes a family of putative nucleotide-binding proteins important for host defense. Although nucleotide binding is thought to be central to this family, this aspect is largely unstudied. The CATERPILLER protein cryopyrin/NALP3 regulates IL-1beta processing by assembling the multimeric inflammasome complex. Mutations within the exon encoding the nucleotide-...
متن کاملThe Relationship between NALP3 and Autoinflammatory Syndromes
The nucleotide-binding domain, leucine-rich repeat/pyrin domain-containing-3 (NALP3) inflammasome, which is required for synthesis of interleukin-1β, has been implicated in the pathogenesis of several autoinflammatory syndromes. This review of the literature summarizes the interconnectedness of NALP3 inflammasome with some of these disorders. Familial Mediterranean fever results from a mutation...

Introduction CINCA syndrome (chronic, infantile, neurological, cutaneous, articular syndrome) also known as neonatal-onset multisystem inflammatory disease (NOMID) represents the most severe form of cryopyrin-associated periodic syndrome ( CAPS). This condition was found to be associated with missense mutations in the CIAS1/NALP3/ PYPAF gene, which encodes cryopyrin. Cryopyrin is a member of th...
